{"id":134413,"date":"2020-02-20T21:12:45","date_gmt":"2020-02-20T20:12:45","guid":{"rendered":"http:\/\/www.madrimasd.org\/blogs\/biocienciatecnologia\/?p=134413"},"modified":"2020-03-06T12:50:27","modified_gmt":"2020-03-06T11:50:27","slug":"nanoparticulas-contra-la-inflamacion-cerebral","status":"publish","type":"post","link":"https:\/\/www.madrimasd.org\/blogs\/biocienciatecnologia\/2020\/02\/20\/134413","title":{"rendered":"Nanopart\u00edculas contra la inflamaci\u00f3n cerebral"},"content":{"rendered":"<p>Un grupo de investigadores de la <a href=\"https:\/\/en.wikipedia.org\/wiki\/University_of_Pennsylvania\">Universidad de Pennsylvania<\/a>, teniendo como primer autor\u00a0al espa\u00f1ol <a href=\"https:\/\/www.linkedin.com\/in\/%C3%B3scar-marcos-contreras-12893610\/en\">Oscar Marcos-Contreras<\/a>, ha publicado\u00a0en la prestigiosa\u00a0<a href=\"https:\/\/www.pnas.org\/content\/117\/7\/3405\/tab-article-info\">revista\u00a0<em>PNAS<\/em><\/a> un\u00a0trabajo en el que han demostrado la eficacia del uso de nanopart\u00edculas en el tratamiento e imagen diagn\u00f3stica de la inflamaci\u00f3n cerebral en\u00a0modelos animales.<\/p>\n<p>La <a href=\"https:\/\/medlineplus.gov\/spanish\/braindiseases.html\">inflamaci\u00f3n cerebral<\/a> aguda ocurre en m\u00faltiples enfermedades como el ictus, el derrame cerebral, el edema cerebral o infecciones causantes de meningitis. Estas patolog\u00edas que progresan r\u00e1pidamente y precisan un tratamiento inmediato,\u00a0afectan habitualmente\u00a0a pacientes fr\u00e1giles, pluripatol\u00f3gicos y de edad avanzada, lo que complica su tratamiento. Los f\u00e1rmacos convencionales apenas se acumulan en el cerebro, requiriendo el empleo de altas dosis e incrementando el riesgo de sufrir efectos adversos. En el estudio de Marcos-Contreras y sus colaboradores, se sugiere\u00a0el <a href=\"https:\/\/es.wikipedia.org\/wiki\/Nanopart%C3%ADcula\">uso de nanopart\u00edculas<\/a> dirigidas a la inflamaci\u00f3n vascular cerebral. De esta forma, con dosis significativamente menores, aumentar\u00eda\u00a0la disponibilidad del f\u00e1rmaco en los tejidos cerebrales da\u00f1ados, disminuyendo los efectos secundarios.<\/p>\n<p><a miden unos 100 nanometros de di\u00e1metro, el mismo tama\u00f1o que muchos virus y unas 50 veces inferiores al tama\u00f1o de las c\u00e9lulas rojas sangu\u00edneas. Las m\u00e1s utilizadas son las conocidas como <\/span><\/a><a href=\"https:\/\/biologia.laguia2000.com\/biotecnologia\/liposomas\">liposomas<\/a><a href=\"https:\/\/nacionfarma.com\/nanoparticulas-diagnostico-patologias-mexico\/\"><span style=\"color: #333333;\">, que son como unas bolsitas de grasa parecidas a las membranas celulares formadas por una bicapa fundamentalmente\u00a0fosfolip\u00eddica. Estos transportadores\u00a0se pueden rellenar con distintos f\u00e1rmacos (mol\u00e9culas peque\u00f1as, prote\u00ednas terap\u00e9uticas o \u00e1cidos nucleicos como el DNA o el RNA). Tambi\u00e9n\u00a0se pueden marcar radioactivamente, lo que permite detectar su localizaci\u00f3n en el cuerpo y utilizarlos para diagn\u00f3stico por imagen molecular. Adem\u00e1s, la superficie de los liposomas se puede decorar con anticuerpos (prote\u00ednas que reconocen de un modo espec\u00edfico otras mol\u00e9culas).<\/span><\/a><\/p>\n<p>En este trabajo, los investigadores marcaron la superficie con un anticuerpo que reconoce <a href=\"https:\/\/en.wikipedia.org\/wiki\/VCAM-1\">la mol\u00e9cula VCAM<\/a>, que se sobreexpresa en los vasos de los tejidos inflamados, dirigiendo las nanopart\u00edculas a la vasculatura cerebral inflamada. Cuando compararon la cantidad de estas nanopart\u00edculas dirigidas contra VCAM\u00a0con las no dirigidas, o con la mol\u00e9cula dirigida frente al receptor de transferrina (una mol\u00e9cula de referencia), se obtuvo\u00a0una acumulaci\u00f3n 300, y 30 veces superior, respectivamente. Gracias a su alta concentraci\u00f3n en el tejido patol\u00f3gico pudieron demostrar\u00a0por imagen molecular la acumulaci\u00f3n de las nanopart\u00edculas en el cerebro inflamado.<\/p>\n<p>El f\u00e1rmaco seleccionado fue un f\u00e1rmaco que act\u00faa sobre las c\u00e9lulas endoteliales, es un RNA mensajero que codificaba la prote\u00edna trombomodulina, que\u00a0tiene funciones anti-inflamatorias y anticoagulantes. Este RNA se encapsul\u00f3 en las nanopart\u00edculas. Al administrar estas nanopart\u00edculas con RNA se logra la expresi\u00f3n de la prote\u00edna de inter\u00e9s de un modo local transitorio. De este modo, administrando\u00a0nanopart\u00edculas dirigidas hacia la vasculatura inflamada transportando un RNA que codifica la trombomodulina\u00a0fueron capaces de disminuir el edema cerebral en ratones.<\/p>\n<p>Como conclusi\u00f3n, podemos constatar la utilidad de dirigir los liposomas frente a ligandos como VCAM y aumentar en gran medida la acumulaci\u00f3n cerebral de las nanopart\u00edculas, especialmente en las regiones inflamadas del cerebro, colaborando con el control de la patolog\u00eda.
