Stein naci\u00f3 en Amberes, de familia jud\u00eda, el 31 de enero de 1931, y tras la invasi\u00f3n nazi, su familia se vi\u00f3 obligada a emigrar a Estados Unidos, en 1940. Stein estudi\u00f3 en la prestigiosa escuela Stuyvesant de Nueva York, marchando luego a Chicago para estudiar matem\u00e1ticas. En esa universidad obtuvo su doctorado en 1955 bajo la direcci\u00f3n de un famoso matem\u00e1tico, Antoni Zygmund.
